Do I have to learn it by heart in order to practice it?

No u don't need to , it's more important that the sound comes out from your Hara center by knowing how to breath while you're chanting ,also when you chant with a teacher , you can learn which sound pitch and frequency to use by simply listening, it's a bit like singing in harmony . .
