Sir, for saying Vanakkam, had observed that few people say "vanakkam" and few say "vanakkangal". Could u explain what's the difference between them and how to use as per sentence? Also as similar to above, when do the words end with "-gal" ?

vanakkam is both casual and formal . While vanakkangaL is very formal where you wish to many people or on behalf of many . I pay my respects to the people here . Here it becomes "respects", is it not?...Likewise it is used in a very formal context. All the best Ujwal

Sir, in this video at 6:25 minutes, "Unge ellarayom paathadhule rombe sandhosham", is it also acceptable to say, "ungE ellarukkE paath rombe sandhosham"

There is no such word as ellarukkE ... you can instead say ungalE paaThadhulE sandhOsho.m = it is pleasure meeting seeing You. ungE ellaarayo.m makes it clearly plural = you all don;t worry you will get there. some little mistakes do happen .
