A good way of finding the Michaelis constant and the (half) maximum velocity of a certain enzyme reaction is to work with a linearized version of your data. Excel can help you to do so.

Hi, thank you very much for the information. I have a question, in the lab we did an expriment with competitive inhibitor and the Vmax should not be affected and Km increasing. However, in my case, the Vmax was decreased and Km increased. I don't know why is this happening.. do you have some ideas? thank you.
