2024 Introduction to Biophysical Techniques
The LMB Flow Cytometry Facility is a core laboratory for all LMB researchers to use and gives the ability to characterise, quantify and separate cell populations on the basis of surface and intracellular markers.
In this video Pier Andrée Penttilä, describes what is flow cytometry, its historical background and uses. She explains how laminar flow is formed and the hydrodynamic parameters that effect it, as quantified by the Reynold’s number. She describes the two main types of sorting systems which use either jet-in-air or a quartz cuvette. In the experimental set-up she discusses the issues of buffer choice, nozzle size for different cell types and the different sort modes to maximise: yield; purity; or single cell isolation. In deciding which type of experiment to undertake she stresses the important of thinking about the downstream processes.
