can I challenge a character in a location or do I have to challenge the location and banish it to get to the character in that location?

unless stated otherwise (like Tiana's Palace), you can target any character as normal even if they are on a location

So when you say “pay the ink cost to move” does that mean an ink is discarded to move to the location?

not discarded, just exerted like if you were playing a character.
