Can you scan in a design and make a stamp from it?

When I get to the last step, my fill comes in but the actual design is not in it. What am I doing incorrectly?

Did you open a design in Embroidery and create the stamp for My Design Center. Without resizing the design, did you choose Add and go to My Design Center, Stamps and establish a boundary for your fill using a shape or the hoop outlines. Place the region fill on the outside of your stamp then choose Set. Because you chose Add before going to My Design Center, your embroidery design should still be there.
