Yasemin is Kosem's little sister, whom we see as a child in the first episode. I don't know where you heard her call Safiye "mother", but if she did, it was probably the term "Valide Sultan", the title used for mothers of reigning sultans. In Safiye's case, she is Sultan Ahmet's grandmother, who was exiled to the Old Palace for betraying him and the dynasty. Kosem had Yasemin killed because she was a spy sent by Safiye to poison Ahmet, so Safiye could finally put her son, Ahmet's uncle, on the throne, and get rid of Kosem and her sons. Safiye hates Kosem, so she purposefully chose her sister to go against her, without Kosem even knowing it was her sister. When Kosem finds out and tries to save Yasemin, it's too late. She has to live with the fact she killed her own blood and flesh.
