99 percent sure they are not related. In the comics Yelena was actually trained side by side with Nat, and even felt jealous of Nat, and along with Melina, was even assigned to kill her, but later they made up. Besides, even if Alexei does kinda feel like her dad, in the comics she was married to him, so until they go completely away from the source material, they are not family.
