1. There is much faster way to do the ROI using the ROI Manager. Also, for mean intensity values, the ROI does not need to be the exact same size in all images, so maybe you could draw a new one for each image. 2. Doesn't the intensity measurement only need to be done in the green channel, not all three channels? 3. What about accounting for the background fluorescence?

This video was not helpful to me at all, plus you have a lot of background noise that you have to clear up in that picture. You do not show a graph, a plot or anything. This video is useless. If anyone needs to see a on plotting intensity look at "Mrs Alicerita" video she goes into detail, shows you tips and tricks and shows you how to build the actual graph not this nonsense
