This being said, I still struggle with the massive dosage that many healhcare influencers recommend for this (180 - 300 mg daily).... I understand that studies shows that there seems to have a benefit at any doses without signifiant side effects, the benefit being higher at higher doses ,,,, very cool indeed and sounds like a major potential breakthrough... but man ... sounds a bit like the far west at this point in time from a clinical perspective as the long term effect of high doses are not known ... I got one dose of immunotherapy, took egcg and metformin (to block glutamine and glucose) and the immunotherapy I got (keytruda) obliterated the cancer in my liver. This was all under the supervision of an oncologist that sayed it was all nonsense. My new oncologist, thank the lord, wants to know how this happened. I also credit my use of probitiotics. Here's a ChatGPT summary: - Melatonin can reverse the Warburg effect, a metabolic process used by cancer cells for rapid ATP production, by redirecting glucose oxidation to the mitochondria. - Melatonin inhibits cancer cell growth by downregulating pyruvate dehydrogenase kinase, which is crucial for converting pyruvate to acetyl-CoA in mitochondria. - Melatonin's ability to switch glucose oxidation from the cytosol to the mitochondria can resensitize tumors resistant to conventional chemotherapies. - Melatonin can convert disease cells from aerobic glycolysis to mitochondrial oxidative phosphorylation, reducing cancer cell proliferation and metastasis. - Melatonin production is crucial at night, and its suppression by light exposure can lead to continuous cancer cell growth. - Exogenous melatonin supplementation is recommended in cancer cases where endogenous production is impaired. - Melatonin reduces cell motility and viability in hepatocellular carcinoma by modulating lactate dehydrogenase activity and reducing glucose uptake. - Melatonin blocks glucose uptake and lactate production, disrupting the Warburg effect and promoting normal cell metabolism. - Melatonin inhibits HIF1 alpha stabilization, a key driver of the Warburg effect, and blocks multiple cancer growth pathways. - Melatonin synergizes with conventional cancer therapies, enhancing their effectiveness. - Urinary melatonin levels can be measured to assess melatonin production and its impact on disease prevention and treatment. - Main message: Melatonin plays a crucial role in inhibiting cancer cell metabolism and growth by reversing the Warburg effect and enhancing the effectiveness of cancer therapies.

Melatonin is actually an immunomodulator (like VitD) as well as an extremely powerful antioxidant (in high doses). It's best to take as a suppository (200mg) vs taking orally which has poor abortion due to first pass metabolism. It also protects against radiation (in high doses) and helps with oxidative stress/excess ROS involved with heavy metals and NPs/MPs (nanoplastics/microplastics), among a litany of other benefits.

Oh boy, a lot packed into this talk. The one thing that stood out that so many don’t talk about and therefore know, is antioxidants affect on cancer cells. Antioxidants are great for healthy cell protection but once you have cancer, antioxidants actually keep cancer alive because of its very aggressive cell division and thus large amounts of oxidative stress that require antioxidants to survive This is actually its weakness or one of, and antioxidants help it survive this which is why you much not supplement which antioxidants or elevate glutathione naturally

The skin has many layers of dead skin cells that provide water proofing. Those cells wouldn't make D because they are dead. Does that help?

My understanding is that Some cancer cells may develop resistance to the effects of melatonin, either by downregulating melatonin receptors or adapting their metabolism. Thus, even if melatonin is increased in the microenvironment of a tumor cell through NIR stimulation for instance , it may not significantly affect some cancer cells. What is your opinion on that Doc ?
@DrCaseyPeavler3 ай бұрын
Remember cancer cells cannot make melatonin due to Warburg metabolism, lack of Acetyl-CoA to synthesize it. The down regulated receptors I am not sure about. But melatonin has both MT receptor dependent and independent mechanisms so that would not nullify its functions.

Speaking of sleep, I just listened once again to Jack on Cameron Borg, and realized how Stasha Gominak teaching about paralysis depth in slow wave sleep is so important, and now Jack talking with Cameron at one point brings this whole points up again in a quantum sense about the purpose of sleep. And, how darkness and quiet is so very functional to allowing the biophoton communication and energy driving physiology. And, the disruptions of NIR starvation, LED light, and non native EMF pollution is so devastating. Melanin, melatonin, NIR, and coherent domain water w/o deuterium in mitochondria needs the deuterium presence in blood for vast flow of light energy within physiology.
@DrCaseyPeavler2 ай бұрын
It’s a light show as Jack says. Some of that light show is absorbed from solar exposure and through non-linear optics transferred to the body systems for utilization. Jack talks about holding that energy and information in water networks and electron excitation. The darkness and quiet at night allows for the next phase of the light show to begin from within. Beautiful.

@@p_peja Early morning (first two hours after dawn), is a great time to get near infrared light (outside in the LIGHT) for a half hour. this helps turn off the pineal gland melatonin production that tends toward sleepiness, and the early morning NIR starts up the mitochondrial production through the whole body (and does not induce sleepiness). This syncs up the whole hormonal process to be TIMMED right! If your mom avoids LED light in the evening, and sleeps in total darkness, goes out each morning to catch the first daylight, she will probably not even need to try to induce sleepiness with taking any melatonin a all, because her pineal-derived melatonin would be enough IMHO.Someone with cancer issues might however benefit greatly by higher doses, as SO WELL suggested by all of these research papers.

To answer the first question, I think that better describes prevention. Healthy weight, healthy diet w/ preserved insulin sensitivity, active lifestyle, perfect circadian rhythm, as much natural light as possible both direct and indirect. So, what sort of dosage of Melatonin are we talking? 1mg? 10mg? 100mg? 500mg? Some suggest we should be taking in the hundreds of mg to get the REAL benefit of melatonin from an anti--aging point of view. The sleep aspect becomes almost irrelevant.
@DrCaseyPeavler3 ай бұрын
Context is king. For non-cancer patients, likely not needed. What is needed is radical changes to your light and dark environment. For cancer, that is its own thing, the dosages regimens are vast depending on integrative doctor and the discussion would ultimately be between a patient and his or her doctor.

As I understood from this video - in high doses it can be pro-oxidative, not antioxidant, correct? What would be unsafe amounts of melatonin to take, upper limit? Approximately. Also, I saw somewhere else that blood levels of melatonin (which u can increase thru supplementation) is not necessarily a direct representation of intra-cellular levels of melatonin, which is what important. Can you please explain what is correspondence between blood levels and cellular uptake.
@DrCaseyPeavler2 ай бұрын
I do not believe it takes high doses. It literally seems to have antioxdant effects in normal cells and pro-oxidant effects in diseased cells. Also has pro-autophagy/mitophagy in normal cells, and blocks autophagy/mitophagy in cancer cells. Its impossibly to clinically differentiate intracellular and serum melatonin production and excretion. We just dont have the tools. Serum melatonin can interact with cell surface MT receptors, can be internalized and act upon organelles such as mitochondria and interact and signal directly in the nucleus.
@jamesthompson72822 ай бұрын
@DrCaseyPeavler That's sort-of helpful, but dodges the question. What kind of dose are we talking about? Supplements offer 3-5 mg typically. Are you thinking that would suffice, or are you thinking one or two orders of magnitude more?

I cannot give specific doses online as this could come off as medical advice or practicing medicine over the internet. What I would say is that the specific discussions about dosing would completely depend on context. Context of patient's health, disease risk and burden. This is best suitable for a discussion between a patient/client and a like minded well read healthcare professional. Discuss risk vs benefit of any intervention proposed and implemented. Follow up and monitoring. My main goal here is to raise awareness, provide scientific backing for these important modalities. I hope that brings context to my at times vague answers.

The question is, what will happen, when you try to get off the Melatonin supplement and normalize your body. Your body got used to the supplement and thus suppressed it's own production and it might be very difficult to bring the body's production back up to normal level. Similar thing happens to body builders. When they get off the supplement (because they cannot continue for the reason of damaging side effects or for financial reason, because these things are costly) they "shrink" like unplugged balloon. So the second question is: does the prolonged use of supplemental Melatonin have adverse effect on the human body? Does it shut off body's own production only temporary or permanently. Is the cancer patient going to need to use the drug for rest of the life. What if it is found later that there are indeed damaging side effect and patient cannot continue (is there any study done about it?).
@DrCaseyPeavler2 ай бұрын
First off, fantastic questions and concerns. They are concerns that my mentor Dr. Jack Kruse and myself share. Most hormones have feedback loops. According to Dr. Russel Reiter, melatonin does not. Therefore you do not shut down endogenous production by taking exogenous melatonin. I think taking melatonin under any other circumstance than cancer is not the smart play. No cancer, maximize melatonin production and release by mastering your solar exposure and darkness at night. With cancer, risk vs benefit, as with all things discussion between you and your doctor.

@@DrCaseyPeavler I watched a few of Dr. Chris Knobbe's videos, where he explains that excess omega 6s in the diet creates leakage in the electron transport chain in mitochondria.

You are awesome Dr. How much melatonin and when should it be taken?
@DrCaseyPeavler3 ай бұрын
Thank you for your kindness and support. Ideally you first and foremost maximize endogenous production. The oral doses for example in cancer treatment would be a discussion between you and your healthcare provider.

Thank you for this. I never knew cancer cells could be fixed. Is that what you are saying? Mitochondria that were ghost mitochondria and non functional can now use pyruvate and perform oxidative phosphorylation with pyruvate as the start compound?
@DrCaseyPeavlerАй бұрын
There is a continuum of mitochondrial damage but in general mitochondrial are nearly irreparably damaged in cancer. If they can be salvaged, melatonin can do it, but if not, then melatonin will induce apoptosis, programmed cell death.

@@DrCaseyPeavler Please do not neglect to include Methylene Blue to help repair mitochondria! Personally I have felt the difference when using it but I am not sure of possible counter indications of use with supplements. Your thoughts would be appreciated.

Ideally inside of your mitochondria and pineal gland first and foremost. Supplemental melatonin is widely available although dosing would be between you and your healthcare provider

Thanks Dr. How can people get sun exposure and avoid skin cancer?
@DrCaseyPeavler3 ай бұрын
Simply don’t burn. Chronic sun exposure is associated with a roughly 15% decrease chance of developing melanoma (the most deadly skin cancer) and an almost negligible increase risk of basal cell and squamous cell skin cancer. When you look at the epidemiological data for cancer deaths those 3 combined make up WAY less than 1%. What people are actually dying from Breast, Colon, Lung, are all decimated by sun exposure. Just take it slow but get out there as much as you can.

@@riumudamc4686 Use sunlight in the mornings and evenings when the infrared part of sunlight that reaches your body is higher. Also look into Methylene Blue for UV damage protection.

Hmmm 🤔 that is a good idea. I’ve never thought about talking about which brands I like…I guess I’m just fearful of them saying I am practicing medicine over the internet. I cannot put my license at risk. Just sharing this information is probably going to put some degree of a target on my back. The channel is mostly to raise awareness and educate. Add one more person praying for you! God bless

What are the modalities of the press pulse that can damage the mitochondria besides metformin ? does HBOT do the same for healthy cells ?
@DrCaseyPeavler3 ай бұрын
That is a great question. I mean I guess what all does press pulse entail? There are many different ideas of what it could look like. But if you use strictly Dr. Seyfried’s framework, then metfromin would be the only obvious one. HBOT likely does cause a mild oxidative stress to normal cells also. But this may act as a hormetic stressor and overall boost health in process. I personally would not use HBOT unless I had cancer.

If you mean like large human trials of high dose melatonin, not that I am aware of. Unfortunately the cost of a study like that would be millions and no one would likely ever fund it. Sadly.
