@Microbehunter2 жыл бұрын
This (unfortunately) does not work. DIC requires not only special objectives, but also prisms, filters etc. etc. and only high-end microscopes can be upgraded to do this. And it is expensive. However, there is a very attractive alternative, called Oblique Illumination, which also produces the 3D effect by creating shadows, similar to DIC. You might also want to experiment with Rheinberg and Darkfield. Video here: kzbin.info/www/bejne/hpzYZYduqJJjosk

I am very new to microscopy, so might be a silly question. How do you get colour on your slides, is it natural? I don't get that at all.

I would be interested in a sample series from varying depths of established soil. A surface, 3", 6", and 12" sample and see how they vary. There is a lot of argument in the gardening world about tilling or not tilling and the theories about disturbing fungal and bacterial strata in topsoil. I personally don't give it much stock, I think whatever upset you cause is re-stratified in a month or so but that's just my hunch.

One more question, Oliver. You said that your camera also captures a very large field of view, therefore there is very little empty magnification. What do you mean by empty magnification? Surely, the field of view can only be as large as the field of view through your microscope objective and eyepiece lenses. I should add that I know about sensor sizes and how they work. I know much more than I need to about still photography, but not so much about videography.
@Microbehunter2 жыл бұрын
When the camera only takes an image from the center of the field of view (and not the whole), then this image is magnified "artificially". It will appear blurry. It is magnification without additional information (beyond the resolution of the microscope) and this is then called "empty magnification". So this is magnification that does not add "value". You can also get empty magnification by using very strong eyepieces. It will appear larger but more blurry. In case of mobile phone cameras, they catch the full field of view, but the image will appear in a circle and you lose pixels this way. You have to crop out a rectangle inside the circle and therefore you again lose some image information.
@nightstormoutdoors60802 жыл бұрын
@@Microbehunter: I have seen this in practice. Thank you for the reply. I do like to keep the circle or at least enough of it to see the diameter of the field of view as seen through the microscope lenses. I almost always use my 10x eyepieces. Putting that circle up on a laptop screen allows me to roughly calculate the size of the specimen by measuring it in mm in relation to the diameter of the field of view, then converting those sizes to microns. If I crop to a rectangle, like you said, I lose more pixels and also any chance of getting measurements of microbes or blood cells accurate. Measuring sizes of specimen is one of my priorities and indeed pleasures. If I keep the circle, it doesn't matter how many times it is zoomed or magnified, the scale of everything within the circle is constant. On video, I usually zoom in to around 2.5 times on my phone camera, this leaves the edges of the circle visible enough to take a measurement across the diameter on the computer screen. So far, I have been very happy with my still images but always looking for ways to make improvements, as there is always room for improvement. I do have a video software program and it can adjust contrast and sharpness, but it is not anywhere near as complex as your software is. My phone camera actually has 13mps not 5 as I stated in my previous comment.

I am a bit confused about your photography techniques. I remember in an earlier video of yours, you said that 5 mps can be even more than enough for microscopy, yet you use a Canon DSLR with far more mps than 5 yet your videos are absolutely crystal clear and pin sharp. How can this be? How can I produce images like that with a 5mp phone camera?
@Microbehunter2 жыл бұрын
Full HD video (1080p) has about 2 megapixel. I use 4K, which has about 8 Megapixels. This is more than the microscope can deliver and the 4K resolution is much higher than what a normal monitor is able to display anyway (and youtube videos are shown small). So it is not a resolution issue alone. the reason why the images appear so clear is because of several reasons: First I have a DIC microscope, which makes the specimen appear to cast a shadow, and increases contrast. So the edges are more visible and you get the subjective impression that the image is clear. Second, I do a lot of editing of the original footage. I use the free video editor daVinci Resolve (can be downloaded), which allows me to adjust pretty much anything. The software is powerful but complex, however. The camera also captures a very large field of view, so there is very little "empty" magnification. So there are several factors that play a role. You will be surprised what simple contrast enhancement is able to do to improve the image quality. While no image processing technique is able to do miracles, I would first do a contrast enhancement (levels adjustment).
