If you use LED illumination, the CRI if your LEDs is extremely important. Older low-CRI LEDs make any specimen look like crap

LEDs can vary widely in color rendition index. One nice thing about halogen is that you're getting the most accurate colors.

Thanks for the in-depth info bought myself a AmScope t-490a custom and I’m loving the quality and sharpness of the image. Thanks again for the helpful advice. Happy microbe hunting.

Great video. I find that with Kohler illumination, I see practically no difference in the position of the iris as long as it is opened wider than the field of view. However, overall, it helps you keep the light source centered and defocused. It is also necessary if you want to add Phase Contrast system. You must have the light source as well centered as possible, I like the LED over the Halogen, simply because it is brighter, allowing you to take photographs at minimum ISO speed. This is extremely important because the noise graininess increases very fast even at ISO 400. This is especially important with still pictures. Videos tend to somewhat cancel out some graininess because the grain speckles are different from frame to frame.

I will need to buy a microscope soon - so thanks for all the videos, helped a lot. What I understood is, that the objecties are pretty much the same in low cost to medium quality microscopes. But what I know e.g. from PCs is that it is not only the compounds but the configuration and how they work together which makes the difference. So isn't it also about the whole setup or the way all the compounds are put together and are aligned? Just thinking... I would need to differntiate between soil organisms. Bacteria, fungi, protozoa and nematodes.

Oliver, in January 2018 I found ur channel... and I first watched ur video on "how to choose a microscope" ....then I decided to buy at that instant... to buy a school level introductory microscope... I have almost completed 2 and half years with it...and have gained a lot of experience from my mistakes... Do u think.. after this much time. Should I advance in my hobby by buying a new microscope like ur Olympus... And if yes...than should I buy it like ur old model Olympus ...or the new ones available in market.. like Olympus Cx33 or Cx43... Please help.. God bless u.

He is saying kohler illumination - found on better microscopes, not so much on cheaper scopes.

As a kid, my first microscope was 50x-900x. I could never focus properly above 450x. Always go for quality, not magnification.

Sorry for posting similar comments under your videos, but I have just seen several of your videos where you give buying advice for microscopes. I wonder why the difference between brightfield and darkfield microscopes doesn‘t have any part in this, not even in this video, where you list lots and lots of differences between microscopes without mentioning this. Isn‘t this something to talk about? There are a lot of microscopes out there that only offer brightfield microscopy, while I‘d guess that a lot of beginners (like myself) are searching for the easier to use and much more versatile microscopes that can do darkfield.

i have recently seem some stereo microscopes with a single eyepiece. It makes some big difference?
@MicrobehunterMicroscopy
@MicrobehunterMicroscopy 4 жыл бұрын
The whole point of stereo microscopy is to get a 3D view. If it only has a single eyepiece, then it looks like a strong magnifying glass. You can of course do amateur microscopy with that as well, and it will have its uses (otherwise they won't have manufactured it) but if you have ever looked through a "real" stereo microscope in comparison, then you will not want to switch back.

What is an Abbe condenser
@MicrobehunterMicroscopy
@MicrobehunterMicroscopy 4 жыл бұрын
It focuses the light from the lamp on the objective. It contains a diaphragm that is used to control depth of field and contrast. Condensers are important for improving image quality. They also have a filter holder.

Which microscope is best for microscopie monocular or binocular
@MicrobehunterMicroscopy
@MicrobehunterMicroscopy 4 жыл бұрын
Binocular for two reasons: A. It is easier to observe over long time by looking through with both eyes and B. many binocular microscopes (these days) are of better quality (monocular are often children microscopes, but not all). Do not confuse binocular microscopes with stereo microscopes! Compound microscopes can be monocular and binocular (1 or 2 eyepieces). Stereo microscopes are always 2 eyepeices. They are for looking at different things.

In the low-cost microscopes they are still common. For high end microscopes (Olympus, Nikon, Zeiss, Leica) the standard is gone long ago...
