Yes, he can support more than 01 application at the same time. But if you have seen this video carefully then you would not waste your time with friend connection points. Even now a days it’s too much challenging to qualify with first blood relative Points as well. So, applying with Friend's point is juat wastage of time and effort. So, if that discourses you.

1 PR holder cannot support 2 applicants at the same time (Fact 4) but what if it is one after the other with time difference of say 3 months in between these two supports. Is it till the 1st applicant receives PR or till MPNP grants 600 CRS points ? As in How long the support or endorsement is stuck with 1 applicant.

If you can satisfy that, then you may expect that. *All candidates indicating that they plan to settle outside of Winnipeg MUST at the time of application satisfactorily demonstrate that they have a convincing connection to a region outside of the capital city indicating a strong likelihood that they will make a long-term economic contribution to that region."

If he and your mom are sibling, then surely he is your close relative. You need to provide both his and your mom's Birth certificates to prove the relationship. Also his Canadian PR/ Citizenship document and his utility bills / rental agreements to prove he is living in Manitoba. Your contribution is very little even if you move to somewhere in Manitoba, your brother will have to have the certain eligibilities as required by the Manitoba PNP. Also, you can support your brother from Regina as well as SINP also acts in the same way when it comes to relative sponsorship. In fact Saskatchewan PNP(SINP) is usually faster than Manitoba PNP. I STRONGLY recommend NOT to move then, relatives can assist a little bit Only, they will have to qualify in terms of IELTS, education and Work experience anyway as per the demand list of Manitoba PNP.
