It was a cool prop, but the is exactly only one way for that prop to work. P&T are clever enough to have worked it out instantly, and not be fooled - which is the point of the show ; )

Since the card in the bottle was straight and flat, it must have been inserted thru a slot that is thinner than the width of the pills, next to the glass side under the pills. Hence we did not see the card until the pills scattered. When the bottle was broken, it was probably shattered along the slot, making it hard to find.

Between 1 and 5 leaves you with 3 options, he has prepared #2, #3 and #4 to palm on second reveal, card fold up in 4 different ways.

If I ask you to pick reds or blacks and you choose blacks, then I say - Good choice, let's eliminate all the black cards. Same goes for diamonds and hearts. That with a 4 way gimmicked card... no FU trophy for that.

I think they learned these tricks from the show. From other Spaniards. As for me, I didn't see any move, only the placement of the card into the bottle, but I had no idea what was happening it's the weak point of the trick, if flawlessness matters.

He had the A-5 of diamonds ahead of time. He said pick a number 1-5. She said 3 so he loads the 3. He asks red or black. Penn says red conveniently, so he stuck with it. If he had said black, he could have said so black is out which leaves red. Same thing with heart or diamonds. It's possible he could have tried a different method but, this is one I've seen multiple times.

...However when they do Fool them, they not only get a FU Trophy, but they get to perform in Pen and Teller's Vegas Show. It can be a major stepping stone for passionate individuals looking to further solidify their career in the magic industry. So while fooling the regular non-magic folks like you and me is nice, fooling Pen & Teller would probably have been more important in that moment.

You should ALWAYS change your answer, yet no one ever does. Are people worried that they'll seem indecisive? You should also never stay stop just as the magician shows down; either say it immediately or wait way longer than seems comfortable ;). "Say stop" is almost never free choice and I despise it.

Each episode is put together from different shows on different days. They don't want all the Foolers to occur on one episode, or an episode to have no Foolers at all. So they cut and paste different performances from different days to make one, balanced episode. That's why Brooke has to wear the same outfit, for the sake of editing shows together.
