Excellent. One thing to mention is that however, p53 does not go by knudson two-hit hypothesis. It is a tetrameric protein, meaning it has 4 subunits. A mutation in one allele may lead to the combination of the single unit with wild type (normal p53 units) or all 4 units being the mutated form. I think this is crucial to mention. Normal tumour suppressor genes (TSG) require mutations in most alleles... But this is slightly different from p53. Great video regardless.

Yep, I was going to mention this error too. If one of the p53 alleles is mutated, p53 function is effectively lost for that cell. Its basic statistics, in this case there is only a 1/16 chance for the wild type subunits to combine to form a wild type tetramer and 15/16 chance a tetramer will have a mutated subunit. 1 mutated subunit is enough for the p53 tetramer to lose its wild type function. Also cells with 1 wild type and 1 mutated allele often lose that wild type allele due to loss of heterozygosity mutations (two hit hypothesis) very readily. In terms of p21, its binding (along with similar protein p27) with CDK4/6::cyclin D actually activates CDK4/6::cyclin D activity which hypophosphorylates pRb allowing the cell cycle to progress from G0/early G1 to late G1 as pRb dissociates from E2F. E2F then activates CDK2::cyclin E activity. The concentration of CDK4/6::cyclin D and CDK2::cyclin E is crucial at this point. p21 (along with p27) inhibits CDK2::cyclin E activity. CDK2::cyclin E is crucial in cell cycle progression because this complex hyperphosphorylates pRb pushing the cell cycle past the restriction point towards the S stage. Once its past the restriction point, it will complete the cell cycle (though it may temporarily halt at the other checkpoints). If enough CDK4/6::cyclin D is produced and binds to p21 (and p27), it will decrease the amount that is available to bind to CDK2::cyclin E.
