Rob Lowe said on the Parks and Rec recap podcast called Parks and Recollection that that “stop pooping” line was when he knew he wanted to stay on the show for the long haul. They originally brought Rob and Adam Scott on for a 6 episode trial run to kind of see if it was a fit. Rob spoke at length during the recap of this episode how he thinks this episode was Parks at peak Parks. To have the stop pooping line and “network connectivity” line come from the same episode is honestly insane. But he said his “stop pooping” line and the laughs it got from everyone around the room and how much they loved it was when he knew the show, set, writers, show runners, cast, everyone were a special group. Good thing too. The version of Parks with Chris and Ben is the version of the show everyone remembers and thinks back on the most I think. It felt like the end of Season 2 and into the Harvest Festival arc of Season 3 is when this show was just off to the races. For my money, this ensemble cast is the best ever assembled in comedy.

Stop pooping and particularly network connectivity problems were both improvs and they’re both iconic lines for their characters. Mike Schur has famously said the network connectivity line honestly made him and the writing staff mad that they’ve never thought of a joke that good lol. I can see why Rob remembers this episode so fondly. Iconic. I could rant about Parks all day, but honestly the cast AND the writing staff was just insane!

improvised doesn't necessarily mean it was the first take of it. a lot of famous "improvised" lines are actually just something that was thought up on the day of filming, but they were made to repeat it because it was deemed funny enough.
