I don't think so, not with just redstone. A comparator only tests for the number of books in it. Command blocks could probably test for what's where, but not what's what. So maybe not even those unless the exact right books are used.

what's fun about these is that not only do comparitors output based on the Last interacted position (and NOT relevant to total books inside), but also the fact that hoppers both fill from 1 to 6, and then take them out 1 to 6...meaning that using a hopper to put them all in will provide the same comparitor outputs as one taking them all out
