l learned [d] sound vanishes when existing between two consonants, however the pronunciation of "husband's room" here is shown as [hʌz|bən|dz rum]; I'm confused about whether [d] of "husband's" should drop or not ?

Merriam Webster defines assimilation (in phonetics) = change of a sound in speech so that it becomes identical with or similar to a neighboring sound the usual assimilation of \z\ to \sh\ in the phrase his shoe. In the VirtuAule System, this element is identified with a dash (ie h ɪ z - ʃ uː )

Hello after some days I have to take a test in phonology one of questions will be "transcript this text in connected speech" my question is do I have to put the stress in each polysyllabic word and connect them together in phonetic symbols ?

+Jetlum Gashi Hello Jetlum - thank you for your question. You should mark the primary stress syllable ( ' ) and secondary stress if present ( ˌ ). The connecting symbol that I use in this video is not a universal phonetic device. I invented it to show students about connected speech.

+Anthony Lombardi Thank you for your answer. So when I transcript the text I have to put the primary stress to all polysyllabic words but not to those with one syllable. and try to connect them together (of course putting brackets (just two of them) )
