Question regarding a desktop set up. Why does it still say no signal when I plug the HDMI directly to my main desktop monitor ? It works when I connect it directly in the computer HDMI port but I need that one port for my main screen.

I think you can't connect monitors to each other - only computers and monitors. There are cheap splitter devices which show same signal on two monitors from one computer, but if you want two different signals then it only works from USB-C with such devices: amzn.to/3YcauiW (affiliate link)

You can do it in Windows using "Calibrate the screen for pen or touch input". Click "Setup" and follow the instructions to select the correct screen.

open control panel and search for tablet PC Settings. You'll need to configure the touch on the correct screen.

I saw a review with this same screen and it was on a Mac. In my opinion it looked even better than Windows 11
