Great question. This is a wet mount, which helps suspend the specimen in place between the slide and coverslip. With the water, it can also prevent air bubbles from showing up on the slide.

Great Video! I am interested to know the difference between the epidermis of a dried vs fresh leaf. Do you have any information about that?

If you conduct a peel with a fresh leaf, you will most likely get live cells. A dried leaf is much more fragile, and its less likely there will be any live cells to experiment on.

Baby Yoda Good question! In a peel, you’re transferring actual plant matter and cells onto a slide. This is useful for experiments. The cells from an epidermal peel will respond to different treatments such as light, darkness, potassium, ABA, etc. An impression only gets the shape of the cells and isn’t useful for behavioral experiments.

@@Ali-rb2gs Yes. A peel is featured in this video. An impression uses another media - nail polish or tape - to transfer the impression onto a slide.
