As someone who has already performed multiple TLC experiments, I feel that this video will definitely help students who want to watch how to perform this experiment before actually doing it in class. This has the upside of the student focusing on the understanding part instead of focusing on how to simply perform the experiment. I also appreciate that this youtube channel highly recommends watching an in-depth video of TLC before watching how to perform it in lab.

Yep, your prediction is correct. We're learning TLC and CC in Orgo Lab rn at my uni and I wanted to watch how to do it so I could understand what was happening.

In addition, if you wish to reuse a capillary you can dip it in acetone to clean it between spottings

Thermofischer is my favorite chemicals supplier. In my lab setting i always order from them, they provide the highest quality amongst the different chemicals that i work with.

TLC can be used quantatively by using different concentrations of the same standard, and comparing the sample spot size to the standards.

Excellent, but you should make many examples for differnt sustances,polar or non polar,in addtion you can explain column chromat. and HPLC.

But why in thin layer chromatography the sample spot to be separated is placed on the TLC just above the level of solvents. Why sir??

If it was initially touching the solvents, then instead of traveling up the TLC plate, the spots would instead dissolve into the solvent and you would lose the spots. If it was above the solvent level, then as the solvent travels up the TLC plate, the solvent would instead carry the spots through the TLC plate, where the spots would then interact with the polar surface of the plate resulting in a slower/faster travel time depending on whether the spots are polar or non-polar.

What's the purpose of capping the solvent and letting it sit, before introducing the plate?

Letting the tlc tank become saturated with the solvent vapour means that the solvent doesn’t evaporate from the leading edge as it rises up. This slows down the rate of movement and the components of the mixture separate into better defined spots rather than streaks. There are some times when this isn’t needed, so you can run the separation without tank equilibration. Also, many of the solvent mixes used are not “nice” chemicals to have in the air…
