I'd recomend looking out for sterile technique (gloves, desinfecting the slides, turn on flowhood, etc..) while slide preping to not introduce false positives.

I hastily bought a couple of microscopes thinking that I could easily see mycelium - I wanted party to do some experiments with my Son but I don't know what clean mycelium looks like and I can't find anything online that is definitive - I've found contaminates but its still pretty hard to differentiate between the two - even with snowy white clean looking samples, its still difficult. Can you show what a clean plate should look like?

What should it look like? Everything you pointed out was the bad stuff. What does the mycelium look like under the scope?

Can you put a syringe filter on the tube, them put that tube into larger container with desiccant.

What if our understanding of fungi is based and spores can connect a third hypae before it finishes merging into one?

some species do this it’s called Di-Mon mating and is spontaneous in nature but can happen in vitro as well.

So you prepared your slide with a non-sterile pipette? You have no idea if the contam was from the pipette or the LC.

Whatever you do, never ever drop a thin slide on your carpet…. 😂 If u do, vacuum very very VERY well.

this is a screening method - I use other methods if I suspect contam, or for more sensitive reasons. there are other screening methods as well so at the time of writing this I have used this a lot but also have used pcr, gram stain procedures, culturing, spiking samples with antibiotics and selective medias and gold standard plating methods all for various reasons (this might make a good video soon)

I don't understand why you love concave slides so much, all it does is make the sample less flat and therefore less easy to focus on.
