I feel like what happened in their past, is what caused Agatha to go seek out the dark hold. Maybe similar to Wanda? Using the dark hold to find her son but then failed due to the dark hold corrupting her.

"you can't kill me" I think means that Lady Death can't kill her, but she can definitely hurt her as we saw at the end "death by a thousand pieces of glass" and the cut on her heel. Death can't take a life, which makes sense because her job is to collect the already dead. Agatha can't absorb Lady Death's magic because Death is ancient and too powerful, for Agatha to even try would kill her. Also, these are breakable rules, witches CAN kill other witches, they are just not supposed to. Agatha killed many witches and many of those witches tried to kill her.
