I agree with many of the other comments -- the content here is made exceptionally clear. Example, no one ever explains the damn name: Major Histocompatibility Complex -- in terms of graft/ host. Your body is using MHGs to check whether the tissue (histo) is compatible with you/SELF. This is but ONE function. He explains this, and also how only the macrophages, dendritic and B cells possess the MHC2 receptor -- its all made so clear in a way that keeps looping back to the overall picture of the immune system. Congratulate Shomu, and yourself for finding him! He seems to really love the subject and this is contagious.

Loading of antigen onto MHC class 2 does not occur in the ER and does not require the proteasome, however loading onto MHC class 1 does require a proteasome and does occur in the ER but it is not for pathogens that have been engulfed from the extra cellular environment, that mechanism is for pathogens that replicate within a cell and thus there is no need for an uptake stage during antigen processing. Please correct that mistake.

Clip replacement occurs in the cytosol and it occurs with the help of HLA-DM. It does not occur freely and randomly as explained in the video.
