A beautifully coherent explanation of a topic that befuddles many. I struggled with XRAY Crystallography for a while mainly since multiple lecturers seemed to assume and ignore the WHY and significance part of the whole endeavour. This overview not only details what each spot entails but creates governable correlations relating Electron Density to the probable structure. Bravo!

Such a good and thought provoking lecture. You are right when you say that the other videos go into the mathematical formulae behind x ray diffraction rather than discussion on the fundamentals. What do you mean by high density of electrons = more chance of finding the fragment of the protein in that specific plane? In a crystal, there are multiple proteins within the crystal packed in a very orderly and aligned manner. So, like do you mean, at that specific plane, a high intensity of the spot = finding a portion of one protein?
