Bro imidazole buffer me imidazole k conc jada h jab hum ye add karenge tab buffer k imidazole groups competition karenge nickel se binding karne k liye aur histidine tag protien(jo bind h nickel k sath) rlz ho jayega(imidazole grp replace the histidine tagged protien)
