Notice how one of the cutscenes shows Rod watching Daniel J. playing with his dad, the youth and happiness Rod never had. I believe this is why Rod targeted him in the first game as his diary mentions he would specifically go to the place where he lived. And although he ended up capturing Charlie instead, he began to hate J. even more when he found out he had rescued Charlie during the events of the first game. Also the way the potion is described here reminds me of the serum from Dr. Jekyll and Mr. Hyde.
