To answer your question about vampires growing back body parts I don't think they can but if their hand gets cut off then the wound left will heal at vampire speed.

Season 7 finally answers the chopping off limbs question. A vampire who gets their limbs cut off and simply reattach it. It was shown when Bonnie chopped off Enzo's hand with an axe.

Exactly Sofie when they turn on their humanity does not approach it denies their mentality because the pain that overwhelms them due to the guilt and remorse they feel is too strong so they tend to focus on those who hate the fact now Elena, Stefan in season 3 and Damon in the season 2 unlike that Damon has been without humanity for more than 50 years and embracing his humanity has been a longer and harder journey.
