I have to say the level of devastation and "I give up completely" is strongest in the scenes when he loses Cas in s12 and s15. Like. Usually he's angry and pumped on adrenaline and wants to immediately fix it. But when he loses Cas its like the pain and heartbreak completely paralyse him. I felt those moments so hard and Jensens acting is always so on point.

I think the scene with Mary should be included in this list. When he tells her he loves her and hates her, how he had to be a brother and a father to Sam. That scene is so full circle and heart wrenching because it pretty much sums up all of his feelings and the fact that Mary's decisions lead to what happened to them. Or when Dean finds out his dad died because he sacrificed himself in order to save him.

When they lose Charlie to the Steines... As much as he blamed them and Sam, it's written all over his face that he blamed himself possibly the most, as she was doing research to help him....
