The most underrated MCAT study channel on KZbin, you do a great job of focusing on the important points, and giving us the "big picture" so we can see where all of these things fit in. I also like that you will continually repeat the overall process instead of getting too hung up in the little details that don't really help with understanding of the mechanism. The GOAT, seriously....

Hello Hassan, thanks for the view! Here is a link en.m.wikipedia.org/wiki/RNA_splicing#Biochemical_mechanism My understanding is that the splice signals are within the introns, as shown in the link.. Also, an Exon can be spliced out if it was between 2 introns that were spliced out... for example imagine Exon 1 - Intron 1 - Exon 2 - Intron 2 - Exon 3 - Intron 3 - Exon 4 - Intron 4 - Exon 5 Then imagine splicing this into Exon 1 - Intron 1 - Exon 2 - Exon 4 - Intron 4 - Exon 5 So in this case, Exon 3 has been spliced out... I’m not 100% confident about the mutation question... perhaps mutations in the introns can affect this process, although this is purely speculative. One thing I can say with confidence is that the MCAT will only ask for big picture concepts, however they do an outstanding job in really making sure you deeply understand those big picture concepts and can apply them during novel contexts. Any esoteric and highly detailed questions will have their answer somewhere in the passage.

I thought exons can be spliced out during alternative splicing to create different isoforms and introns can never be in the final mRNA. Alternative splicing comes from different mRNA exon combinations and the introns are always spliced out. Is that correct?

Are spliceosome sites located on the introns? Also how can a mutation in an intron sequence cause an exon to be omitted from the final mRNA transcript? (question from one of the AAMC tests). Great video
