I figure many will already know this, but here's a quick breakdown anyway: in Japan, blood types are often informally used as indicators of personality, somewhat like a Myers-Briggs test. A blood types are considered well-organised perfectionists who tend to be stubborn. This is why at 1:02, they wonder if chat is A type since they comment about the sea lantern balance. Botan is pretty chill and easygoing, so she doesn't fit the traditional A blood type conception, hence why they are surprised. B blood types are creative but they are also selfish, not team-oriented, and may be hard to get along with. Meanwhile, O types are outgoing and chill. I personally think Botan would 'fit' this O group. I can see why she guesses Pekora might be an O type at 3:14, since they are also known for their leadership skills (and Pekora is the shachou after all).

O type usually portrayed by great social skill, a social butterfly kind of person. while B usually portrayed as antithesis of A type who often seen as perfectionist, B usually seen as easy going and my own pace type of person, even though they're flexible most of cases, they tend to do their thing haphazardly, kind of egoistic and not a team oriented type of person, "this is fine" meme perfectly portrayed B type. Since Japan is kinda workaholic kind of society, there's a view that B type is not good at their job that need team effort, while A type often nitpick other's job.
