Butters was based off the creators friend, if I remember correctly in an interview, they were saying their friend would actually say "gee fellas"

Butters is based on Eric Stough. The South Park animation director and producer. Trey Parker and Matt stone based the character on him.

And Kenny was based off an old friend of theirs. Who always wore a huge orange coat and mumbled all the time.. And he would always miss days of school, so they started saying he died as a joke every time he didn't show up

Yes, some characters are based on people the creators knew. Liane Cartman was named after Trey Parker's ex-fiancé who cheated on him; so he named Cartman's promiscuous mom after her as a dig at her.
