i've found that by having my lab bench a little bit higher it helps take some of the strain off my back when working, would highly recommend it!

I cool off my scalpel with a quick dip in a small glass that has 70% alcohol in it. Also, when saving mycelium from a contam'd dish I do that work inside a still air box rather than in front of a fan so I'm not blowing spores all over myself and the room.

The thing i like most about having plates on hand (my case jars) .. The condensation built up mostly evaporates by the time you use it...

Hi, very nice video, can you explain if it's possible to transport some mycelium from other countries (UE- USA ) to nord Africa, I want to make a small mushroom farm using mycelium of lion's man ...etc witch are not known in my country, do they have a lifespan ? Thanks for sharing, super informative! Lab skills and process are really important and theres not a whole lot out there to help with the learning curve. very timely for me as I have been spending alot of time in the lab lately refining my process, finally working on backing up my cultures. Funny note to leave you with....I was pouring plates the other day and wouldn't ya know it the mason jar I was using slips out my hand and of course it pours hot agar on/into my flow hood, so the lesson that day...get a handle of things. Simple yet so valuable.

oh man, that sounds like a nightmare. I once dropped a glass jar of spawn on my lab floor when it was in my spare bedroom back in 2013, and had to remove all the shelves and sweep up the mess. It was a lot of work and frustrating.

Hey man just curious I thought it was bad practice to so agar transfers where one is infected with mold in front of a laminar flow hood because the air from the flowhood will cause the mold spores to blow all over the room in which you did the transfer leading to even more contamination. I thought in this instance you would be better suited to use a SAB?

good videos I see them from colombia south america and I want to put the translation option because I am very interested in what you say, I also have a mycelium laboratory in my house and I have not been able to get even a petri dish with mycelium mother to inoculate grain . the boxes with culture medium always begin to sweat that is normal? with or without inoculum sweat I do not know why I cover them with parafilm and that always happens to them, that I will be doing badly.

I am not going to say this wasn't a very informative video. I do a ton of agar work myself and one of the number one rules is NEVER open a contaminated dish in front of a flow Hood. I'm sure you can understand the science behind it. The mold spores that are in there will begin to flow around everywhere. Still air box work for contaminants only. At least that's my company's rule of thumb. Please don't think I am trying to BASH you. Just some constructive feedback. But I'm sure you know enough to know what you need to know. Great video!

Is there an update video on the contaminated transfer? Did it work out? Did you need to transfer again? For really bad infections how many transfers do you think it could take to isolate? New to agar work. Thanks for your patience. Where are the vents on the petri dishes you recommend, honestly I am a bit confused about the concepts of vents in petri dishes in general? I know they need to breathe but aren't the lids on them already loose? I didn't notice you covering any vents with tape. Do the boxes of 500 come in sleeves? Are they sterile?

Another great video. Curious about where you buy the plastic jars you use? Looks like you put on the filter disks yourself. There's always something satisfying about the time lapses, and you always use a certain type of music that I've come to associate with WTF. Funky jazz reminds me of Let's Grow Mushrooms and It's neat to see the different styles reflect different personalities.

when i pour agar infront of a flow hood, do i need to leave the cabinet running until it solidifies and i can wrap it with parafilm? im used to a sab but i just spent a grand on a nice hood, but i dont know about when the agar is still warm and liquid and you just poured it.

Hello. Very interesting cinnamon and quite instructive! I have a few questions. Would you share your experience with us? 1. Have you tried putting agar or wheat mycelium in a chamber. minus degrees in order to stay longer. What are the results? 2. Can you move from agar to agar after a certain period of time. As far as I understand, there are up to about 4 times the grain after which the qualities are lost. For example, after 2 or 3 months, move to a new agar? 3. How long can the mycelium persist and what are the steps? Thanks in advance!

Yes it can. 3% peroxide will not kill mycelium however

although it will kill spores! So no peroxide with Spore work. Only with mycelium work. There are actually agar recipes that contain peroxide :-) and it is very effective I might add from personal experience

@@whatthefungus I get that the filter hood will handle the threat just fine. But still feel like that hood just spread green death over everything down flow of the hood. Might be an interesting experiment with some open plates behind a contaminated plate behind a hood, see what comes of it.

I would rather get process, protocol details versus an inventory of your cultures. NTK: temp, humidity, duration , light level, transfer techniques, how to spot contaminants, sources for tools, supplies, costs, sources of inoculants, etc.

In my readings about soil health everyone talks about how fungi works symbiotically with other organisms. You patureize your growing medium to kill everything much like practicing monoculture in farming - not healthy and reduces nutrient density in crops. Please comment.
