I don't think it will matter either way. Might be easier to install the tack strip if the glass is out... you do have to ben some of those metal tabs.

They make non marring plastic removal tools

Actually the proper way to remove the trim that’s installed in the rubber is to remove the glass first then remove it from the rubber, that way your chances of damaging the trim is way less.

Just purchased a 53 210 two door this evening....It hasn’t been driven since 1962 and it has just over 50,000 original miles. But it sat outside for all those years…..It has a little bit of rust on the top corners of the windshield ....This video will help when it comes to taking off the trim around the windshield!
