In this tutorial, I cover a simple method of scattering particles on an animated character, and binding them so the particles follow the movement of the character's animation. We then take it a step further, and interpolate procedural splines among the neighbor particles, and simulate physics on the resulting splines in a secondary flow. The resulting splines are bound to the original scattered particles, and are affected by gravity and motion physics of the animated character.

Is it possible to use Tyflow to simulate fat/muscles simulation on a character? can you tell me how? I'm using cloth modifier to simulate jiggle and deformation on fat, but I'm sire it could be better with Tyflow

Is it possible to simulate chain replacing the splines.

Yes, sort of. You can specify a mesh in the tysplines object, and it will conform to the splines as they drape and move around. Basically, you can model a series of chain links in a straight line, make them a single object, and specify you chain object in the tysplines object.
