Is it possible that it will be positive for any other body fluids or secretion rather than semen?

There are low levels of acid phosphatase found in other body fluids like vaginal fluid, so it may produce a color change, but not as strong as what is seen in semen.

Hello. I tried Acid Phophatase test on different body fluids for my study. Semen turns purple in ~20s with high concentration of semen but I was surprised to see that other body fluids make the test purple very quickly also : leap mucus in less than 3mn, saliva en around 5mns, eye swears in 7/8mns and even human sweat in around 10mns. So can it be considered as reliable test as it seems many other body fluids contain acid phosphatase and can react quickly (of course with lighter purple compared to semen based on what I saw) or at least it seems to be very difficult to interprete a result (My teacher told me also that several vegetable produce fully false positive such as tea). Does such test needs to be confirmed with other test as it seems highly possible to have false positive (or wrong interpretation dpeending on the time the stain will start to become purple). Hello and thank you so much for your question! The Acid Phosphatase test is presumptive and labs do not report out positive results of this test. They only report it out when it is negative on a surface stain. Semen is confirmed with either a sperm search or a the presence of P30. I have not seen all the cross reactions that you got. Our main cross reaction has been with Vaginal fluid.

@@dawonfields7360 Acid Phosphatase is an enzyme found in high concentrations in human semen. It is secreted from the prostate gland and cleaves phosphate group from molecules. It is the basis behind the Acid Phosphatase test which is a presumptive test for semen.

According to our forensic scientists, anything that contains acid phosphatase should come back positive. So in theory, yes, but more research would need to be done to answer this definitively.
