I also have videos on the Gram Stain, Flagella Stain, Endospore Stain, Capsule Stain and more if you want to check them out! Here is a link to a playlist on all my videos relating to Microbiology Stains:
• How to Pass Your Micro...
Biology Professor (Twitter: @DrWhitneyHolden) explains the difference between acidic and basic stains (dyes), a handy pneumonic device to help with memorization, and the uses of these stains in microbiology. Stain structure is also discussed, including chromophore and counterion descriptions and examples of multiple stains are given.

Is acidic stain different from acid-fast stain? Slightly confused here. Thank you xx
@BiologyProfessor
@BiologyProfessor 4 жыл бұрын
Yes, they are different. Acidic stains are dyes that are negatively charged (like the bacterial cell walls) so the dye is repelled And gives color to the background the cells are on rather than color to the cells themselves. On the other hand, the acid-fast stain is a staining technique that allows one to differentiate between acid-fast cells and non-acid-fast cells. I have a video on the acid-fast stain here if you are interested: kzbin.info/www/bejne/inSpoYhupZh1r5Y

Ma'am why proteins are stained by acidic dyes and nucleic acids by basis dyes ?
@BiologyProfessor
@BiologyProfessor 6 жыл бұрын
Acidic dyes react with cationic (positively-charged, basic) components in cells. Proteins fall into this category (they have a lot of amino acids with positive charges), and so they will bind to acidic dyes. Basic dyes react with anionic (negatively-charged, acidic) components in cells. Nucleic acids are acidic, and therefore bind to basic dyes. You can find more information here: histology.leeds.ac.uk/what-is-histology/H_and_E.php
