This is because the main guitarist lost four fingers of one hand playing bowling, after that they let him make that tone at the beginning of each song as something symbolic for all that he had contributed to the group.

Today, producers have "producer tags" they put at the beginning of rap songs as their trademark. The Ink Spots simply pioneered that idea, this riff is their "trademark"

I don't think it's really supposed to be an intro, just something they played on the guitar to get their timing right (like how drummers do "tap tap tap" before a song begins.)
