Aseptic Technique: Inoculating a Petri Plate - Streaking for Isolation.

This video lesson will demonstrate how to streak a Petri plate so as to obtain some single isolated colonies after incubation. It also shows how to remove bacteria from a Petri plate.

Hi Lili,The key is to keep the entire loop wire almost parallel to the agar. If you hold the plate at eye level and lift the lid just enough to insert the wire it forces you to keep the loop at the proper angle for good streaking. If the plate is on the bench top and you lift the lid too high the tendency is to point the loop downward where it digs into the agar instead of gliding over the surface.
