Basic Plant Tissue Culture Part 1

The purpose of Basic Plant Tissue Culture video produced by Professor Acram Taji is to provide the theoretical as well as practical basis of plant tissue culture for the production of higher plants via in vitro techniques. The video covers the following important areas:
Stock plant management
Media preparation
Aseptic procedures
Stages of micropropagation including initiation, multiplication, rooting and acclimatization (hardening off).
This video is aimed at researchers, teachers, students, nursery people and other individuals who are interested in plant cell, tissue and organ culture, particularly in clonal propagation.

Generally pretty good, but I take issue with two things. At one point, the worker dips a piece of pH paper into the media, which is potentially a source of contamination. It probably won't make a difference, but the better way to do that is to dip a clean stirring rod into the media, and then touch the stirring rod to the pH paper. The more serious mistake was when the technician touched the 0.22 uM filter with his bare hand to insert it into the plastic filter cassette. Assume that your hands are full of bacteria. Since both sides of the filter are now assumed to be contaminated, bacteria will get into the solution after it passes through the filter. This should be done instead using a sterilized set of forceps, or better yet, buy pre-sterilized disposable filter cassettes to attach to syringes. Probably you'll get away without problems most of the time, but why risk it? The best single strategy in tissue culture is paranoia.

@@drpk6514 If you're doing filter sterilization, it's because the component (eg. acetosyringone) is heat labile, and has to be added after the media has been autoclaved and cooled. Hands in general are considered sources of contamination, so a good working rule is that your hands never touch the experiment. This is also a good safety practice. For the same reason, there is no justification for dipping pH paper directly into any solution. Probably the amount of chemical leached from the paper into the solution is not going to matter, but why add another unknown to the experiment when there is a cleaner way to do it?

@@brianfristensky521 There is absolutely no problem with using pH meter in the media. We are dealing with plants and they are not all that sensitive. The filter which was used for the filtering has 2 protective layers, the protective layers were placed on the hand to show the filter to the camera. Furthermore, these days no one uses these types of filters. They come in one time use ready to use one piece. BTW that "worker" has a PhD in plant biotechnology and this is his job.

Can one simply take fresh seeds from an aquatic plant (Aponogeton) and germinate, grow them in a nutrient-rich tissue culture gel, even without all of the sterile lab equipment and procedures? I have an abundance of seeds which would be much easier to recruit than slicing and sterilizing bulb chunks to put into a sterilized MS medium.

If you do that then you will have a microbial bloom in the media and soon takes over your seeds/small plants.

@@drpk6514 but orchids are grown from seed in the same way so I'm sure it is possible. Maybe will have to tweak the gel mixture though

@@xPorsum I have worked with Orchid seeds too. If you get them out of capsul you need to sterilise them and if in green capsule, you need to extract them without any contamination.

What is that filter tool called?; the one used at the end of your video for cold filter sterilization it looks like a female nozzle tip for your syringe. I need to sterilize thidiazuron.

Yes they don't sell to individuals easily. I tried to buy something for myself but later they reviewed my purchase and cancelled it. However you can buy pretty much everything from eBay or smaller companies online. You could even buy prepared medias online easily.

I ended up going to Phytotech labs instead. Pretty much have everything you'd need for plant tissue culture.

This is tissue culture so you don't need any pesticide/herbicide.
