Note: The media after autoclaving looks yellow in this video. For demonstration purposes, we used a different media afterwards to save time, however, please keep in mind that MS media does not change color like this while you do this experiment in real.

no, you add whatever required hormones for what your are growing.

If I dissolve all the materials into water and adjust ph than also add agar. But I have no time to autoclave. how many days can keep the media in 4 degree? Or I need to prepare again

It depends on the agar you are using. Some looks more brown.
