While they are mostly machines to us, Cybertronians do have a biology; iirc it was Starscream or Ratchet that referred to their Transformation Cogs as more like organs than mechanical machine parts. As for the pepper spray, it could be that there was a reaction it had with something in Knockout’s optics, be it metal itself or something internal. Or hell, it could have just covered his optics like when you get water on a camera lens, simply for comedic effect.

Knockout is the perfect representation of a cybertron between the sides of the dicepticons and autobots. He isnt heartless or ruthless like starscream or megatron. Yet he isn't all goodie goodie like the bots. Just a guy who lost his home and wants to find a content place in his life. Something megatron and optimus could both respect when it comes to their core values. Unfortunately the two can never come to an agreement as one is too hell bent on being the sole ruler of cybertron and the other just wants to continue on and find new peace in life amongst humans. Seeing knockout at least interact with people unlike the other dicepticons just proves that the cybertronians are no different from humans. Just that two completely apposing powers can't get along with each other.
