The funniest thing about this scene is that the extras playing the soldiers were not told anything about this scene only that they were not allowed to laugh and had to maintain a stern face. So their reactions and laughing is completely organic and Palin is completely chewing up the scene trying to get them to laugh.

the try not to laugh part always makes it harder so when you are force not to laugh then most people fail at not laughting.

That's not true. Palin's film diary describes having to do multiple takes of this scene with the extras and needing to keep coming up with new names for Pilate's wife to try to keep the energy and humor up at the end.

I first saw Life of Brian when it was released in Perth, Western Australia. The US Navy (7th Fleet?) was visiting at the time so the cinema was almost a third full of young sailors and Marines just about the time that Monty Python was being appreciated in the USA. The whole place was roaring with laughter, especially as the Monty Python shorts 'The Christmas Card' and 'Away From It All' were shown beforehand. By the end I'd laughed so much my ribs hurt and I'm sure I wasn't the only one. A happy memory.

As many others have said, the extras playing the guards were told not to laugh at anything, though the reason why doesn't seem like many people know. In order to get the reactions out of the guards that they wanted, they told them they only had the budget for one take and so they absolutely couldn't afford to screw it up and that it's a very important scene to the film. So the guards holding in the laugh is actually them trying their best not to ruin the take.

When the movie was released, the local council of the town where she and husband lived banned the movie from being shown, considering it disgusting. The joke being that Roman Friendship was indistinguishable from Roman Enmity, with the only real difference being it's ironic name.

The speech impediments of very rich Romans was real and also happened to the upper classes of England in 18th 19th and early 20th century. The reason was the same in both cases: piped water; that came through lead pipes, to the homes of wealthy people. This then caused their children in particular, to develop a range of developmental problems; including to the speech centres of the brain. That is why the Toff with a speech impediment became a standing joke in British Music Hall. It is hypothesized to be one of the causes of the fall of the Roman empire and was something the Monty Python team, learned in the history classes of the University where they first met.
