Hi Arpan, I was wondering about when you drew the 3 curves of the CD Spectrum why the alpha helix curve looks like how you drew it, why the beta strands look like how you drew it and the random coils too. If I was given a CD spectrum of my own, I assume I can predict the secondary structure by comparing it to how closely it follows the 3 curves you have drawn, but I wanted to know the underlying principle. Thanks

+saravanaraj m concepts of ORD and CD are quite similar(but they have a fine line of difference) i would say both are manifestation of same underlying phenomenon. spectropolarimeter is based on the principles of ORD .It measures ability of an optically active compound to rotate polarized light as a function of wavelength(one could think it as wavelength dependence of optical rotation).ORD is about the way the material refracts polarized light in a different way, when the wavelength of that light is changed. ORD is measured as the difference in absorption or extent of interaction with the incoming light, when wavelength of the light is changed. ORD is usually used with plane-polarized light, and the change in optical activity is tracked as wavelength is varied. As we saw with CD, ORD examines the wavelength effects of linearly polarized light incoming upon the material, and looks at how much the recorded CD is being changed as different wavelengths are used. Short wavelengths tend to interact more strongly with materials, compared to longer wavelengths, so we experiment with different wavelengths to do an ORD spectrum. What we get out of an ORD spectrum is the difference in transmission speed of left- and right-circularly polarized light going through the material. The way CD and ORD operate is built in such a way that the two methods can provide information about each other. With ORD associated with dispersion and CD connected with absorption, it is possible to use Fourier transforms to convert between these two. If the ORD spectrum shows a positive tendency, then long wavelengths of the polarized light are interacting more strongly with the material, creating a positive CD band. If the ORD spectrum goes in the opposite way, this means a negative CD band, and short wavelengths of polarized light are interacting more with the material.
