To carry out the aseptic technique you firstly use an oven called an autoclave to sterilize any equipment. It uses heat to kill all pathogens to stop cross contamination. You then set the Bunsen burner to a blue flame, which is used to sterilize the inoculating loop, and the air around where you are working. Open the lid of the bacterial culture a little, always working by the blue flame of the Bunsen burner. Dip the inoculating loop into the bacteria and transfer the bacteria to the agar plate. Use the inoculating loop to streak the bacteria across the agar. Sterilize the inoculating loop in the blue flame. This prevents cross contamination of pathogens. Seal the lid, but make sure it isn’t airtight, so anaerobic pathogens don’t grow. Place sealed agar plate into an incubator set at 25 degrees Celsius, so the bacteria can grow, and colonies can form.
