This is an excellent video with excellent analogies that makes it super simple to understand. One extra detail to note here is that Pyruvate Kinase can also be activated by F16BP (+ feedback). I believe this was mentioned in Kaplan MCAT Biochemistry.

It is also high yield to know that conversion of glyceraldehyde 3 phospate into 1,3 biphosphohlycerate needs NAD+ as cofactor of glyceraldehyde 3 phosphate dehydeogenase. This is something that has been asked in uworld in the context of alcohol ketoacidosis where there is NAD depletion (due to NADH acummulation) or LDH deficiency, where you are NAD depleted, as in normal conditios the conversion of pyruvate into lactate uses NADH and finally converts into NAD to be used in this specific glycolisis step. In this 2 scenarios you would not have NAD, thus glycolisis can not procede anymore as there would not be any cofactor for G3P dehydeogenase, explaining why patients with alcoholic ketoacidosis and LDH Deficiency have hypoglycemia.

What I fail to understand here is that under other resources it also lists Glucagon as an inhibitor of Pyruvate Kinase alongside ATP and Citrate which appears counterintuitive. Do they not exist in complete opposite sides in terms of the availability of glucose and energy?

Didn't know that fructose 1,6-bisphosphate gets converted into both DHAP AND glyceraldehyde-3-phosphate. The DHAP gets converted to glyceraldehyde-3-phosphate by triosephosphate isomerase so that means there's TWO glyceraldehyde-3-phosphate per glucose molecule. WHY'S that important? well it explains why glycolysis produces net 2 ATP excluding the NADH produced in the cytosol which needs the malate-aspartate shuttle or the glycerol phosphate shuttle, mitochondria, and oxygen to produce energy. The books say glucokinase/hexokinase and phosphofructokinase uses 2ATP total phosphoglycerate kinase and pyruvate kinase produces 2ATP total that would result in a net 0 ATP IF there was only one glyceraldehyde-3-phosphate. Knowing now that there is two glyceraldehyde-3-phosphate produced we know that 4 ATP is produced which means there's a net 2 ATP produced. Knowing exactly why and how glycolysis produces 2 net ATP in normal circumstances, this explains why RBCs which produce 2,3-BPG bypassing phosphoglycerate kinase when there is severe anemia or hypoxia results in a net 0 ATP from glycolysis. This also makes the ETC video make sense when you explain the net ATP produced from glycolysis, to pyruvate metabolism, to oxidative phosphorylation.
