Hello Mycophiles, in today's video I show you all an experiment that has been in the works over the last year. I have been saving any leftover petris I may have from a pour, to see if I can use them later instead of throwing them away. Was it a success?
I hope you all have a great day or night!

Did anyone else think Petris was a game which involved tessellating bacteria or domestic animals? The title "Don’t Waste Leftover Petris! How to Save Them" I thought it was a game guide. Awesome content as ever.

I had one sleeve with one cracked dish in it after delivery, and fragments of the dish pierced 3 cuts in the sleeve (~ 5-8mm long). I used other dishes without any sanitation (just like regular sleeve) for my transfers and had zero contamination 😄

If you do unmodded lids you dont even need aluminum foil for them. Foil does help with contam though.

I have been able to get multiple uses out of my used single used petris by washing them with soapy water spaying the inside and outside with iso and h202. I close them up and let them dry in my sab. Then I pour them like their new and I have only had a handful of failures.

You can pour far more with a sleeve of petris than you can make prepours, plus you have full visibility + control condensation. You also do not need to clean them after you’re done. There are pros and cons to each.

They're great for a photo contest but not practical for mycology imo. Having to sterilize them every time and lifting up stacks to pour sucks when using glass. Not worth it when I can get a case of 480pcs for 57$.

I have glass plates that I wash with soap after use. I mostly use these. But I also reuse my single use plastic plates, when I do use them. I also just wash them with soap and store them in the sleeve they came in when I bought them. I don’t bother sterilizing any of them. I have a pretty unconventional way to pour plates. I leave them open while they cool in my still-air-box, so I will have less condensation. When they are cool, I close and seal them. I have a strong UV-C light in my box that is turned on while they’re cooling, so I don’t have to worry about the plates being sterile. The light kills all contaminants anyway, while the plates are cooling. I’ve never had a contaminated plate since I started doing it this way. I leave the room while the light is on, because it’s dangerous to the eyes and skin.

@CorteX1988
@CorteX1988 Жыл бұрын
I don't use gloves at all. As long as your technique is good it should be perfectly fine. As for foil, I've recently just been using a whole sheet and laying it on top of my jars while PCing instead of doing them one by one. This prevents any water from landing on the jars and you can just re-use it again as much as you want. This is assuming you have good lids of course.

Not worth it imo, disposable are cheap, no cleaning time involved, no need to sterilize, and doesn’t break. The only downside is environmental unfriendliness, but other than that it wins on everything.

@@spacepirate1543 idk, you also have to clean them, larger so more of a PITA to stack, plus they’re not any more sterile than disposables (assuming they’re sterilized correctly in the first place, whereas with disposables you don’t need to worry about that).

@@Mycophilia you wash them in the dish washer and put them in a grow bag , fold the bag up at the top and remove all air. tape the bag with the blue painters tape it will go through the pc unscathed just chuck them in the cooker with the agar and pull them out in the sab (i have a flow hood so i do that) the staking is the hardest part dude just give it a try also works with Polypropylene ones micropose just started selling that don't break and could theoretically last a life time. but i like glass because it keeps the agar from drying out and can be used for long term storage if properly sealed like a year if you pour them thick enough
