Hi..thank you for this video..may i know if i can substitute the peptone and NaCl with bacteriological peptone?...and can we confirm the presence of e.coli by dropping kovacs reagent on violet colonies? can i know where can i get the documented ISO protocol for this video? or is this validated in house method?

You can use bacteriological peptone. Don't drop kovac's reagent directly. Do it as done in this video. You can purchase the protocol from ISO website

Our laboratory doesn't have stomacher machine, what should we use as a substitution? Is it okay to shake it by hand? Second question is that, how much water sample should we dilute?

You can shake with hand properly to make homogeneous mixture. Dilution 25ml of sample into 225ml diluent to make 10 times Dilution
