Very nice presentation and straight forward procedure. I will soon have 2 undergraduate students try this kit for research projects on salivary exosomes. We'll definitely get in touch soon.

@@CellGS we ordered 2x EX01-25L Exo-spin™ kits (24 columns), unfortunately the kits too long to arrive due to Covid-19 travel restrictions and cold chain was disrupted. Upon receipt, we stored columns and buffer at 4oC but our fridges broke down over Christmas holidays and we only noticed when we reopened. The students had fun with the kits though, procedure was straight forward but the samples did not elute from the column, although we strictly followed low-speed centrifugation steps. I think the functionality of the columns was affected by inappropriate temperatures during transportation and storage. We'd like to try again with this year's group. Thanks for asking

How are exosomes differentiated from viruses?

Not possible! there is no difference between them at all, what they call viruses are actually exosomes! We have been deceived big times The remarkable resemblance between EVs and viruses has caused quite a few problems in the studies focused on the analysis of EVs released during viral infections. Nowadays, it is an almost impossible mission to separate EVs and viruses by means of canonical vesicle isolation methods, such as differential ultracentrifugation, because they are frequently co-pelleted due to their similar dimension [56,57]. To overcome this problem, different studies have proposed the separation of EVs from virus particles by exploiting their different migration velocity in a density gradient or using the presence of specific markers that distinguish viruses from EVs [56,58,59]. Hi Gungun Barman, Exo-spin kits are optimized for isolating exosomes in this size range. Just follow the standard protocol and you'll achieve this.

Only size based separation cant purify from microvessicles and apoptotic bodies...... Then why you called it exosome? It should be EV for clarification.

@@sharatbarman3352 good point, but I don't think there is a kit on the market that would allow you to separate exosomes from smaller ApBs, MVs or even ectosomes. This is why you ALWAYS need to verify by WB your desired ESCRT biomarkers

Hi Mohammad, Yes it's very effective for both plasma and sera. There are larger sizes for bigger samples and a 96 well version for purifying large numbers of samples.

Chemically do exosomes have anything to do with the immune system other than changes in chemistry per say? I don't know if it would work? Would the use of the trillions per second camera combined with electron microscopes be able to see chemistry in us?

Exosomes transport molecules between all cells, including immune cells. Cytokines are transported in exosomes between immune cells to help orchestrate the immune system. There are many techniques that allow the study of exosomes both in-vitro and in-vivo. They can be visualized with electron microscopy.

@@CellGS thanks for your reply, but why? Everything bigger than 30nm will be in a void volume anyway so all EVs together if iam not mistaken :). Also when i will the use precipitanion buffer, in the manual it is written that the polymer in the buffer causes that LC-MS downstream method can not be used. Does it mean that it can not be used only immediately after precipitanion or also desalting on a spin column wouldnt help? Thanks :), we need to use it for the LC-MS method.

@@matusjurcik6974 Good question. The reason larger particles >200nm don't come through is because the space between the resin beads isn't big enough. THe 20-200 nm range is because smaller things soak into the cracks in the individual resin beads and larger things just don't get through the gaps between the resin beads efficeintly. So that creates a window of sizes that are purified dependent on the circumference of the resin beads. If you are using LC-MS downstream, we recommend concentrating the sample with other methods such as a micro concentrator. If your starting sample is blood, you won't need to concentrate. You can just add the sample directly to the column. If you have any more questions, please contact tech@cellgs.com to speak with an expert.

@@CellGS Thanks :), ok so it means that there is only Vvoid + Vpore (small contaminants/proteines) and retarded Vvoid alone because some ways can be blocked by bigger (more than200nm) particles right? because bigger particles will stuck ... Also we try to extract EVs from the plasma to screen biomarkers of deseases and we want to use the LC-MS for that.

@@matusjurcik6974 You're rigth. The bigger particles will eventually block the passage of the target exosomes. That's at least in part why there's a limit to the number of times you can reusue the columns. They get progressively slower. If your using plasma, there's no need to concentrate the sample. For screening biomarkers where you're likely to be using large numbers of samples, Exo-Spin 96 is worth looking at.

Although not recommended and they are economical enough for single use, it is possible if you flush them out with PBS, if you're not too worried about sample carryover.

This product is available from www.cellgs.com and is for research use only. We sell to research institutes, pharma and biotech companies. There is a lot of research on the regenerative potential of exosomes. As with stem cell therapy, there's also hype and nonsense out there. Just using exosomes purified from any source will not work. I'm not aware of any completed clinical trials showing exosomes work therapeutically. Until properly organized trials have been completed, we can't be sure they work.

Hi Stefan, Thank you for your question. This technique may not be how you would go about isolating viruses. Nevertheless, I think it's reasonable to assume that any viruses present in a sample would co-elute with exosomes in an SEC column based on their size. I have no problem with the concept that beneficial viruses exist. But that does not mean pathogenic viruses don't exist. Pathogenic viruses do exist. Vaccines help the body to fight them.

Pathogenic virus organisms exist? Where is the proof of that? Where is the proof that a vaccine stops these highly contagious deadly mutating pathogenic virus organisms from getting inside the body and being pathogenic inside the body? If that was a true statement by you then then those who didn't get a vaccine would be dead of virus organism infection and that's definitely not the case as many people I know have never been vaxxed against any pathogenic virus organism and have no issues living life. Many people have beeng around people who have been told they have highly contagious deadly mutating virus organisms growing by the billions inside their bodies exploding their cells even though they too feel absolutely fine and have been told they are asymptomatic but have lots of virulent highly contagious deadly mutating virus organisms growing inside them but are not sick… Some of these people have been vaxxed and still are told they have these pathogenic virus organisms growing in them so how does that work? Should we all run and hide from these people or accept the fact that the germ theory of disease is a fairytale and so are highly contagious deadly mutating pathogenic virus organisms.

Everything you said was wrong, exosomes and corona are one and the same, exosomes are pieces of a dead cell, the corona, and it can be used as delivery system for the brain, it can carry nano technology, graphene, for brain / machine interface to the quantum computer, with the help of 5g towers.
