in this video I demonstrate an experiment I did with a colonized BRF jar of Pes Amazonian. I made a karo LC with a sterile syringe.

I expand cultures when I get them (Peptone and malt). However, my syringes never go empty anymore either. When it gets down to a few MLs, I simply suck up some fresh lc mix. Syringes will “colonize” much faster than jars of LC. Sometimes I do something very similar to this method above. TIP- you can get extra long blunt tip needles and sharpen them on a fine stone. This is very handy for reaching past the verm in pf jars, pulling LC without gettting liquid near the lid, and for getting all over the place when NOCing up jars or bags. I have a syringe thats over a year old, never been in a fridge. It all started when I got a culture syringe that had a visible chunk (looks like they used blender) that would never come out. If you dont let them sit forever, and use proper sterile methods, it seems like you can keep it going quite a while just giving it more mix.

Could you just use sterilized water from a vacuum sealed container of sterilized water ? I work in the medical field so just curious

So you squirt it in the colonized jar and then extract it back up? Or you just hope you get enough on your needle ?
